FSD Fluor™

FSD Fluor™ is a new generation of dye series with superb fluorescence intensity and high quantum yield comparing to traditional dyes. Flamma® Fluors

BioActs offers a broad range of Flamma® Fluors dyes equipped with variety of reactive and functional groups, which can cover the full spectral range from UV to NIR with their excellent fluorescence performance. Characteristic features of these superior dyes are strong absorption, high fluorescence quantum yield and high photostability. Flamma® dyes maintain good fluorescence activity and stability after conjugation to biomolecules and allow the detection of low-abundance biological structures with great sensitivity. The dyes are compatible with optical conditions of most of fluorescent equipment and are ideal for any applications in biological studies.

ICG Dyes

ICG is clinically approved NIR dye and used in medical diagnostics, in vitro, vivo and animal model study. NIR fluorescence allows to observe the deep image from the surface of skin and its usage spreads to a wide range of research fields. ICG fluorescence method is safe and cost effective technique and used in a wide range of medical fields such as breast cancer sentinel lymph node navigation, detecting cerebral vessels, coronary arteries and biliary trees, tracking tumor location, detecting small HCC, etc. The method has several advantages such as radiation free, compact instrumentation, real-time monitoring, easy operation, etc. The maximum excitation/emission values of ICG are 785/821 nm. When ICG is injected into a human body, it rapidly bound to plasma protein, mainly high-density lipoprotein, and generates red-shifted fluorescence (845 nm). ICG in aqueous solution is unstable over time, thus the fresh solution should be used for effective trials. BioActs provides ICG dye for biological research and medical diagnostics and also offers various reactive and functionalized ICG dyes for labeling of antibodies, peptides, proteins and ligands.

Other Labeling Dyes

BioActs provides other traditionally used dyes such as 5(6)-Carboxyfluorescein (FAM) and 5(6)-Carboxytetramethylrhodamine (TAMRA) dyes for labeling of biomolecules. FAM is one of popular green fluorescent reagents used for labeling peptides, proteins and nucleotides. In addition to relatively high absorptivity, good fluorescence quantum yield and good water solubility, FAM has an excitation maximum that closely matches the 488 nm spectral line of the argon-ion laser. TAMRA fluorophore has been a widely used for preparing bioconjugates, especially fluorescent antibody and avidin derivatives. TAMRA dye is also widely utilized for oligonucleotide labeling and automated DNA sequencing applications. TAMRA is often used as FRET acceptor for FAM fluorophore.

qFlamma® Quenchers

Dark quenchers are substances that absorb excitation energy from a fluorophore and dissipate the energy as heat without any native fluorescence. The combination of a fluorescent dye and quencher may act as an on/off switch of fluorescence that the original fluorescence might be restored as the distance between them increases or when their connection is cleaved. In conjunction with fluorophores, dark quenchers are regarded as key components in designing FRET probes, in protease activity assays, nucleic acid hybridization, PCR, etc. The main advantage of dark quencher is the non-emissive excitation and elimination of background fluorescence. BioActs developed a new generation of azo-anthraquinone derivative dark quenchers, qFlamma® quenchers, to overcome issues associated with existing dark quenchers such as stability, quenching ability and water solubility.

The absorbance peaks of qFlamma® quenchers completely cover spectral range from visible to near-infrared (NIR) that all of commercially available fluorescent dyes can be effectively quenched. The quenchers display high absorption coefficient together with excellent quenching ability, superb stability and aqueous solubility so that it can easily manipulated in aqueous buffers. The four of qFlamma® quenchers (Black 01, Red, Blue and Orange) series cover the electromagnetic spectrum from 400 nm to NIR region and provides excellent spectral overlap across the entire range of known fluorescent dyes. Fluorescent Dyes for Oligonucleotide Synthesis

Fluorescent oligonucleotide probes are developed for the detection and the quantification of target DNA or RNA sequence with the high sensitivity and specificity along with low toxicity. They have been used in a wide range of applications including, FISH, in situ hybridization, sequencing and genotyping. Fluorescent dye and quencher paired molecular beacon probes are widely used in the field of real–time nucleic acid detection and PCR quantification, SNP detection, and clinical diagnostic assays.

There are two major strategies for chemical incorporation of fluorescent dyes into an oligonucleotides: direct fluorescence labeling during chemical synthesis and post synthetic fluorescence labeling. The direct oligonucleotide labeling during solid phase synthesis can be achieved via the standard phosphoramidite chemistry containing various spacer arm lengths. Direct labeling during solid phase chemical synthesis might afford high yield but also increases the risk of damaging fluorescence tag from acidic deprotection step. Thus certain fluorescent dyes are not compatible with solid phase chemical synthesis, thus their labeling must be conducted post–synthetically through various labeling methodologies.

Post synthetic labeling of a fluorescent dye to an oligonucleotide can be achieved by labeling various activated dyes to modified oligomers having functional groups such as primary amine, thiol, aldehyde, azide, alkyne, or carboxylic acids. Depending on binding target, the fluorophore might be selectively introduced at either the 5′ or 3′ ends of the oligonucleotide as well as randomly incorporated throughout the sequence.
