Description
In recent years, many diseases such as cancer, arteriosclerosis and diabetes, and the association with macrophages have been reported in large numbers. In order to examine the relationship between disease and macrophages, it is necessary to conduct experiments such as "what happens if there are no macrophages?" In such cases, this product is a reagent that can efficiently deplete the culture system or macrophages in vivo.
Feature
- This product is a reagent in which clodronic acid, which is a kind of bisphosphonate, is encapsulated in liposome.
- Clodronate, as a competitive inhibitor, inhibits the metabolism of ATP and damages cells.
- However, clodronate alone does not have very high macrophage-depleting activity due to its low blood retention and low cell membrane permeability.
- Liposome formation improves retention in the blood and cell membrane permeability, and the increase in apparent size by liposome formation also enhances the recognition by macrophages.
Outline of Experiment
200 μL of this product was intraperitoneally administered to normal mice, and 24 hours after administration, the liver and kidney were removed for real-time PCR and histopathological staining.
Results
- Real-time PCR
- Tissue immunostaining
