Description
MAdCAM-1 is an immunoglobulin (Ig) cell adhesion molecule family member. MAdCAM-1 is involved in lymphocyte homing to mucosal sites and is expressed on high endothelial venules (HEV) of both mesenteric lymph nodes and Peyer's patches. It has also been found to be expressed on sinus-lining cells of the spleen. The integrin, alpha 4 beta 7, has been shown to function as the MAdCAM-1 receptor. The Ig domains of MAdCAM-1 have been found to be critical to alpha 4 beta 7 binding. The mucin domain has been shown to have activity in L-Selectin binding.

Human Diseases
- Ulcerative Colitis (UC): MAdCAM-1 is upregulated in UC, contributing to leukocyte trafficking and inflammation in the gut mucosa. Monoclonal antibodies targeting MAdCAM-1, such as ontamalimab, have shown efficacy in clinical trials for UC treatment.
- Crohn's Disease: Similar to UC, MAdCAM-1 plays a role in the pathogenesis of Crohn's disease by mediating leukocyte adhesion and migration to inflamed intestinal tissues.
- Multiple Sclerosis (MS): MAdCAM-1 may also be involved in MS due to its expression in the central nervous system during inflammatory responses
Cellular Signaling Pathways
- Integrin Signaling: MAdCAM-1 interacts with the α4β7 integrin on leukocytes, facilitating their migration to mucosal tissues. This interaction is crucial for immune surveillance and response during inflammation.
