Protocol & Usage

Standard Freezing Protocol

  1. Cell Preparation: Harvest cells at 70-80% confluence

  2. Medium Addition: Resuspend cells 1:1 with OptiPresev

  3. Cooling Rate: 1°C/minute using controlled rate freezer or isopropanol bath

  4. Storage: Transfer to liquid nitrogen (-196°C) after reaching -80°C

  5. Thawing: Rapid thaw in 37°C water bath, dilute immediately

Optimization Guidelines

  • Cell density: 1-5 × 106 cells/mL recommended

  • Cooling rate: Adjustable for different cell types

  • Storage duration: Validated for >2 years at -196°C

  • Thaw protocol: Cell-type specific modifications available

Handling Guidelines

  • Maintain sterile aseptic technique throughout use

  • Allow to equilibrate to room temperature before use

  • Mix gently by inversion before aliquoting

* All DiagnoCine Precision Sterile media are filtered-sterilized with a 0.1-micron filtration two times and 0.04 micron once. Thus, mycoplasma contamination is prevented.  The smallest size mycoplasma type can be about 0.2 microns.
