Application Notes
Forphenicine is isolated from fermentation broth of Streptomyces fulvoviridis var. acarbodicus MC974-A5.1,2 Forphenicine shows potent inhibition against alkaline phosphatase prepared from chicken intestine (IC50 value is 0.036 μg/mL) and slightly inhibits the enzymes prepared from calf intestine, liver and Escherichia coli. The type of inhibition of chicken intestine enzyme by forphenicine is uncompetitive with p-nitrophenyl phosphate and its Ki value is 1.64×10-7M.1 Forphenicine enhances delayed-type hypersensitivity (DTH) and increase the number of antibody-forming in mice. Forphenicinol, orally administrable forphenicine derivative, has been studied.3,4
Role in Human Disease
Immunomodulatory Effects: In animal studies, forphenicine has been shown to enhance delayed-type hypersensitivity and increase antibody production capacity. This suggests potential applications in modulating immune responses.
Anti-Tumor Activity: Forphenicine has demonstrated inhibition against Gardner lymphosarcoma in certain studies, indicating possible anti-tumor properties.
Cell Signalling Pathway
Alkaline Phosphatase Inhibition: By inhibiting alkaline phosphatase, forphenicine can potentially affect various signaling pathways that rely on phosphorylation and dephosphorylation cycles. These cycles are essential for activating or deactivating proteins involved in signal transduction.
Impact on Phosphorylation: Since phosphorylation is a key post-translational modification that regulates numerous cellular processes, including metabolism, cell growth, and apoptosis, the inhibition of enzymes like alkaline phosphatase can have downstream effects on these processes.
