A standardized, lyophilized coagulation reagent containing highly purified bovine thrombin, formulated for the quantitative determination of thrombin time (TT) — also known as thrombin clotting time (TCT) — in human citrated plasma. Upon reconstitution and addition to a pre-warmed plasma specimen, thrombin directly converts fibrinogen to fibrin, bypassing all upstream coagulation factors and isolating the terminal step of the common pathway. The clotting time recorded (in seconds) reflects the quantity, quality, and functional integrity of fibrinogen, as well as the presence of thrombin inhibitors such as heparin or direct thrombin inhibitors (DTIs).

The thrombin time assay interrogates the final and most direct step of the coagulation cascade: the thrombin-mediated conversion of soluble fibrinogen to insoluble fibrin. Unlike the PT and APTT, which activate the cascade upstream and require the sequential function of multiple coagulation factors, the TT test bypasses the entire intrinsic, extrinsic, and common pathways — adding a known, standardized concentration of thrombin (Factor IIa) directly to the plasma sample
