ThelioKeep is a serum-free and dimethyl sulfoxide (DMSO)-free medium for preserving for 1 - 2 weeks at 4℃ 

Epithelial and endothelial tissues have various functions in living systems. These tissues are characterized as thin membranous structures, physically fragile and difficult to preserve. ThelioKeep keeps the structural strength and viability of these membranous tissues.

Preparation of ThelioKeep-EGCG

By using a micropipette under the hood, take 0.5 ~ 1.0 mL of ThelioKeep from the bottle, aseptically pour it into the microcentrifuge tube containing EGCG Tablet and completely dissolve the tablet in the tube. Pipette the entire solution back into the bottle of ThelioKeep and mix well (=ThelioKeep-EGCG).

Usage

  • Preservation of epithelial and endothelial tissues
  1. Place a few mL of prepared ThelioKeep-EGCG into a clean disposable tube and cool it at 4 ~ 10 °C.
  2. Immerse epithelial or endothelial tissues freshly isolated from experimental animals into the cooled ThelioKeep-EGCG prepared in (1). Close the cap and preserve it at 4 °C for 1 ~ 2 weeks.
  • Restoring live tissues from preserved state
  1. (1) Gently wash the preserved tissues, indicated above, a few times in cold PBS. Transfer the tissues to an experimental medium.
  2. (2)-1 When the live tissues are used for experiments or cultured, place them under physiological conditions as soon as possible (e.g. 37 °C).
  3. (2)-2 When slide specimens will be prepared, transfer the tissues to fixatives 
  4. (2)-3 When cytoplasmic materials will be extracted from the preserved cells, wash the cells well with cold PBS.
Expiration
The product can be used for one year at 4 ~ 10°C after manufacturing when ThelioKeep and EGCG Tablet are stored separately. 

Storage
The prepared ThelioKeep-EGCG must be used as soon as possible. Left-over solution can be stored at –20 °C for up to 2 weeks without loosing its functional quality. Do not freeze and thaw ThelioKeep-EGCG repeatedly.
Precautions
  • This ThelioKeep-EGCG has not been tested on all epithelial and endothelial tissues. Test the solution on a few samples before you preserve the entire batch of tissues in your experiments. This product is for research use only.
  • In some tissues, the concentration of EGCG in the solution may be too high. In such cases, adjust the amount of the tablet to be dissolved in the solution to reduce the final concentration.
  • We, BioVerde, are not liable to any accidents or damages caused by improper usage of the product.
Quality control
Bacterial tests: 
Bacteria, fungi or mycoplasma
contamination is all negative.

Chemical tests:
(1) pH = 6.7 ~ 8.0 at RT 
(2) Endotoxin < 0.5 EU/mL

Human corneal endothelium tissue were preserved for 2 weeks at 4C.
1-(A) : When using ThelioKeep, hexagonal shape of the cytoplasmic membrane was well maintained.
1-(B) : When using another company's tissue culture medium, cell morphology was completely disappeared.
2 : When ThelioKeep was used, the cell proliferation speed was about twice compared to when another company's tissue culture medium was used.
