Description:
Ponceau-S is a dye for staining proteins with reversal manner. Because Ponceau-S can quickly stain and decolorize proteins, and does not interfere the antibody reactions, it is widely used for verify the protein transfer to membranes in western blot. This product is a ready-to-use Ponceau-S solution for the use of protein-transferred membrane.

How to Use
1.Removal of membrane
Remove protein-transferred membrane from blotting apparatus.
2. Staining
Put the membrane into plastic box, and pour Ponceau-S staining solution into the box so that the membrane is fully soaked. Let it stay for 2 to 5 min while shaking.
3. Decolorization of background
Discard the staining solution from the plastic box, and then add 1% acetic acid solution. While gently shaking, remove excess staining. Normally 2 min is enough to remove background staining, but it is better to examine state of staining by eyes. (We recommend to take pictures at this stage if needed. To do so, remove the membrane to wet filter paper. Be careful not to dry the membrane)
4. Total decolorization
Discard the solution from the plastic box. Then pour 0.1 M NaOH solution (Dissolve 4 g of NaOH to 1 L of DW) so that the membrane is fully soaked. While gently shaking, remove staining. Normally 10 to 20 sec is enough to remove all staining.
5. Washing
Discard 0.1 M NaOH solution, and wash by DW by shaking for 2 min. The membrane is now ready to move the process for antibody-reaction step starting from membrane blocking
