Mazethramycin B (mazethramycin 11-methyl ether) was isolated from culture filtrate of Streptomyces thioluteus ME561-L4. The absolute stereochemistry of mazethramycin B was determined by the X-ray diffraction analysis. Mazethramycin B had antimicrobial and antitumor activity. It prolonged the survival period of mice inoculated with leukemia L-1210 ( T/C: 240% at 31.3 μg/kg/day). Mazethramycin B has the pyrrolobenzodiazepine (PBD) structure, which is a class of natural products produced by various actinomycetes. Pyrrolobenzodiazepines (PBDs) are known as sequence-selective DNA alkylating agents with remarkable antineoplastic activity. Ongoing research demonstrates that PBDs can be effectively used for antibody-targeted therapy.
