Description:
Hektoen Enteric (HE) Agar is a moderately selective, differential medium designed to recover and differentiate Salmonella and Shigella from other enteric flora. The bile salts inhibit most Gram-positive organisms and suppress many Gram-negative commensals. The carbohydrates support acid production by non-target enterics such as coliforms. pH dyes such as bromothymol blue and acid fuchsin shift color to show whether the medium has become acidic from carbohydrate fermentation or alkaline from peptone metabolism. Sugar-fermenting flora acidify the medium and shift the color toward yellow to orange. Peptone metabolism by Salmonella and Shigella alkalinizes the medium, keeping the colonies and surrounding medium green to blue-green. Sodium thiosulfate as a sulfur source and ferric ammonium citrate as an indicator yield a black ferrous sulfide precipitate where H2S is produced. Hektoen is often used in the isolation and differentiation of Salmonella and Shigella from fecal specimens diagnostics. It is also used in food safety and public health labs to test food, water and environmental samples.
CO₂ incubation: CO₂ incubators can acidify the medium and alter pH, so plates should be incubated in ambient air, not in CO₂.
