Human Diseases
- Cancer: Overexpression of MYST4 is linked to various cancers, particularly ovarian high-grade serous carcinomas (HGSCs) and hepatocellular carcinoma (HCC), correlating with poor patient survival rates.
- Developmental Disorders: Mutations in MYST4 are associated with several syndromes, including Noonan syndrome-like disorder, Ohdo syndrome, genitopatellar syndrome, and blepharophimosis-ptosis-epicanthus inversus syndrome.
- Acute Myeloid Leukemia (AML): The MYST4 gene can fuse with the CBP gene due to chromosomal translocation, contributing to AML.
Cellular Signaling Pathways
- RAS/MAPK Pathway: MYST4 regulates this pathway, influencing cellular growth and tumorigenesis.
- Acetylation of Non-Histone Proteins: MYST4 modifies various proteins involved in critical cellular processes, including transcription regulation and DNA damage response.
- T-Cell Signaling: MYST4 plays a role in T-cell activation and function through post-translational modifications that affect signaling cascades.
