Acridine orange is a fluorochrome stain which differentially stains DNA and other cellular structures. It absorbs UV light and emits visible light. DNA is stained yellow green whereas cellular sub-structures including RNA are stained orange.

Acridine Orange is used in detection of sister chromatid exchange during karyotyping and in detection of RNA in DNA.

Quality Control:
  • Appearance: Brown to red - brown or violet powder.
  • Solubility: Clear very dark brown to orange solution at 5gms in 10 ml of water.
  • Dye content: ~ 80%
  • Absorption maximum 1: 489 - 495 nm (0.005 g/l in water)
  • Absorption maximum 2: 285 - 291 nm (0.005 g/l in water)
  • Absorption maximum 3: 265 - 271 nm (0.005 g/l in water)
  • Suitability for electrophoresis: passes test
  • Loss on drying (at 110°C): NMT 5.0%
